Pharmaceutical giant Roche Holding Ltd (RHHBY) has unveiled a pivotal clinical trial plan for its Alzheimer's disease antibody drug, aiming to demonstrate its ability to delay disease progression before symptoms manifest.
As competition intensifies, the company is racing against rivals like Eli Lilly and Eisai to advance Alzheimer's care from a treatment-focused model to a preventative one.
The study, named PrevenTRON, plans to enroll 1,600 participants who show biological markers of Alzheimer's but have not yet developed any cognitive impairments.
The research will utilize Roche's experimental antibody drug, trontinemab, which incorporates Brainshuttle technology designed to deliver a higher and more stable dose of the medication into the brain.
Roche disclosed key details of the trial design at an Alzheimer's Association conference in London this week, including plans to begin participant recruitment later this year.
The company intends to use a blood test to identify individuals with elevated levels of the plasma pTau217 biomarker, which can pinpoint those who appear healthy but are at high risk of developing Alzheimer's symptoms in the coming years.

Eli Lilly has been conducting preventative research with its approved Alzheimer's drug, Kisunla, for approximately four years and is positioned to be among the first to report results from a large-scale preventative trial using an approved anti-amyloid drug.
Similar to Roche, Lilly employs blood tests to screen cognitively normal individuals at the highest risk, identifying them before symptoms arise.
The president of Lilly's neuroscience business, Carole Ho, expressed a vision for a future where patients can be identified via blood tests years before symptoms appear, allowing for early intervention.
This optimism is gaining wider acceptance across the research community.
A senior director at the Banner Alzheimer's Institute noted that anti-amyloid drugs work better the earlier they are administered, making preventative studies like those from Roche and Lilly the logical next step.
An executive from the Alzheimer's Drug Discovery Foundation suggested that Roche's Brainshuttle technology could potentially amplify the effectiveness of this treatment strategy by delivering more medication to the brain.
Instead of a fixed trial duration, Roche will employ an event-driven study design, continuing the trial until a sufficient number of participants develop measurable cognitive impairment.
